We are currently offering a fantastic career opportunity for highly motivated and experienced antibody discovery/development scientists to join Antharis Therapeutics, a pre-clinical company based in San Diego. The company is committed to discovering and developing novel immunotherapeutics (monoclonal antibodies and antibody-drug conjugates) for various disease states, including oncological, neurological, inflammatory, and infectious diseases. The approaches for antibody discovery, including high-throughput screening methods and hybridoma technology, aim to expedite the development of lead candidates for entering the clinic.
The individuals selected for these positions will be responsible for the Company's antibody discovery and development programs. Functions include intellectual input and decision-making on protein targets to be pursued, execution of immunization strategies, utilization of high-throughput screening and hybridoma technology, antibody validation in-vitro and in-vivo, and humanization of assets, all of which aim to expedite the development of lead candidates for entering the clinic.
These positions require strong, highly motivated, and fully independent scientists who will use a hands-on approach on the bench to develop therapeutic antibodies. With guidance from the Company’s CSO and Director of Research & Development, these individuals will lead immunotherapeutic product development efforts and, consequently, will have significant intellectual input in the direction of the asset pipeline. We are seeking a Scientist I with hands-on experience in antibody discovery using hybridoma technology. Responsibilities will include designing and executing immunization and screening strategies to generate panels of antibodies that target various proteins for novel targets in the immuno-oncology, oncology, neurology, auto-immunity, and infectious disease fields. The candidate must have in-depth knowledge of current state-of-the-art antibody generation technologies and be able to apply this to practice at the bench.
Responsibilities:
• Design and execute approaches to immunize mice and perform electrofusion
• Design and execute hybridoma cell culture using both liquid and semi-solid media
• Design and execute screening protocols using both ELISA and flow cytometry methods
• Proactively propose strategies to project teams to generate the target antibodies
• Execute all stages of antibody generation to deliver diverse panels of antibodies meeting the required criteria.
• Integrate data from multiple sources (affinity, epitope mapping by competition binding, target sequences, and initial antibody purification)) to generate a complete data package for antibody characterization.
• Maintain detailed records of all experiments in ELN using in-house databases and tracking systems.
• Direct a group of scientists abroad to facilitate the hybridoma generation workflow
• Responsible for delivering both data analysis packages as well as purified antibodies to project teams
• Troubleshoot and optimize protocols and propose creative solutions to improve the efficiency of existing processes.
• Proactively evaluate new technologies, instrumentation, and assays available from the literature or scientific meetings to improve in-house platforms.
Qualifications:
• Ph.D. and postdoctoral training in Molecular and Cell Biology, Immunology, Biochemistry, or a similar advanced degree with 5+ years of relevant experience.
• Demonstrated experience successfully executing antibody generation using hybridoma approaches to identify antibody panels is required. B-cell cloning is a plus.
• Extensive experience with antibody binding assays and analysis, especially ELISA and multi-color flow cytometry, is required.
• Experience with PCR, molecular cloning, and transfection of mammalian cells is a plus.
• Strong experience in Excel, GraphPad Prism, and flow cytometry analysis software is required
