As part of this role, you will be responsible for developing assays to understand monoclonal antibodies expression systems using non-natural amino acids. Your experience in RNA, DNA, and protein analysis techniques will be valuable in optimizing our non-natural amino acid platform and facilitating the production of high-quality biotherapeutics.
Join our pioneering mass spectrometry laboratory and gain access to the latest instruments, robotic workbenches for automated sample handling, and powerful bioinformatic software and cloud computing resources. Key Responsibilities:
- Develop and implement mass spectrometry assays to optimize protein expression levels, protein quality, and quantitation of nnAA orthogonal elements (such as tRNA and RNA synthetase). Ensure accuracy and success of MS experiments through expertise in sample preparation.
- Use advanced liquid chromatography-mass spectrometry (LC-MS/MS) technologies for protein identification and intra-cellular quantification.
- Employ top-down mass spectrometry methods to quantitate truncated protein both inter- and intra-cellularly.
- Analyze antibodies with nnAA incorporation using the peptide mapping technique to identify the site of insertion and perform sequence variant analysis.
- Collaborate with other functions and teams to design experiments that address critical questions.
- Troubleshoot technical issues and propose innovative solutions to enhance analytical capabilities.
- Stay up to date with the latest advancements in oligonucleotide, protein quantitation, and cell engineering. Develop new methods and publish findings in peer-reviewed journals.
- Prepare and present scientific findings and recommendations to internal customers, project teams, and senior management.
Qualifications
Required:
- Ph. D. in life sciences or a related field with 0-3 years of experience OR a Master's degree with 4-6 years of experience
- Strong background and hands-on experience in oligonucleotide techniques, peptide quantitation, sample preparation, mass spectrometry-based analysis, and interpretation.
- Knowledge of tRNA extraction techniques and mass spectrometry characterization of intracellular protein expression components.
- In-depth experience operating mass spectrometers from all vendors and optimizing instrument parameters and data analysis.
- Proven track record of working independently and collaboratively in a multidisciplinary team environment.
- Excellent problem-solving skills and attention to detail.
- Strong written and oral communication skills, with the ability to effectively summarize and communicate in-depth MS results to multi-disciplinary teams.
- Experience in oligonucleotide mass spectrometry workflows and data analysis.
- Experience working with non-natural amino acids (nnAA)
- Advanced knowledge of protein and genetic components required for expression systems.
- Proven experience in mass spectrometry protein characterization.
- Experience with protein quantitation via Multiple Reaction Monitoring MS (Mass Spectrometry) methods.
- Prior experience in the biopharmaceutical industry or related fields.
- Solid understanding of mass spectrometry and MS sample preparation.
- This role is based in Spring House, PA and may require 5% travel.
